Custom DNA / RNA oligos
DESALTED
Desalting helps to remove most of the low molecular weight by-products that form and accumulate during chemical synthesis reactions. This purification method is suitable for oligonucleotides of less than 30 nt and/or those intended for routine, low-criticality laboratory applications, such as PCR, sequencing, probing, mobility shift, or hybridization. We would like to emphasize that desalted oligonucleotides are not recommended for molecular cloning projects, for which oligonucleotides purified by superior methods, such as HPLC, are advised.
HPLC PURIFICATION
HPLC purification (high-performance liquid chromatography) is recommended for oligos longer than 80 nt and for all more sensitive laboratory procedures. This purification method allows for the removal of truncated oligos and residual products. HPLC is also particularly useful for achieving higher purity, which is necessary for successful applications that require higher sensitivity, such as cloning, DNA fingerprinting, real-time PCR, FISH, etc.
OPC PURIFICATION
This purification method consists of reverse-phase chromatography (RP), which separates the complete, newly synthesized oligonucleotides from defective sequences, by-products, and other impurities. More specifically, the OPC® cartridge binds the dimethoxytrityl (DMT) group of the oligonuclotide carrying the trityl (complete, newly synthesized) from molecules that do not carry the DMT. This technique is particularly effective for oligonucleotides up to 40 nucleotides in length.
